Sequence and origin
Tuftsin (Thr-Lys-Pro-Arg) is a tetrapeptide fragment of immunoglobulin G heavy chains. Selank adds a Pro-Gly-Pro extension that resists C-terminal proteolysis, producing a measurable extension of plasma half-life relative to unmodified tuftsin. The PGP extension also appears to alter receptor pharmacology in some published assays.
Mechanism hypotheses
Several mechanism hypotheses appear in the literature: GABAergic potentiation, BDNF expression modulation, and direct modulation of monoamine turnover. The mechanistic picture is not settled; in-vitro work should pre-specify which hypothesis the assay is testing rather than assume a single canonical mode of action.
